      Background InformationE3 ubiquitin-protein ligase HACE1 (UniProt Q8IYU2; also known as HECT domain and ankyrin repeat-containing E3 ubiquitin-protein ligase 1) is encoded by the HACE1 (also known as KIAA1320) gene (Gene ID 57531) in human. HACE1 is a ubiquitously expressed HECT (Homologous to the E6-AP Carboxyl Terminus) family E3 ligase with relatively higher expression in heart, placenta, kidney, and brain. HACE1 is a 909-amino acid protein composed of six ankyrin (ANK) repeats/domains (a.a. 64-257) in its N-terminal region and a HECT domain in its C-terminal half (a.a. 574-909). HACE1 a dual function E3 ligase that mediates target cargo degradation by at least two independent mechanisms with different E3 ligase requirements. HACE1 participates in proteasomal degradation via its HECT domain E3 ligase and in autophagic degradation via its ANK domain-mediated protein–protein interactions. HACE1-mediated cellular proteostasis only becomes apparent under stress. Much of HACE1 research activities are focused on its roles in oxidative stress in brain, autophagy in the heart during haemodynamic stress, and stress-induced tumorigenesis.
