SpecificityDopamine b-HydroxylaseDBH is a single gene product that exists as a cytosolic enzyme, as a membrane protein and is secreted into the circulation (Lewis & Asnani, 1992). Because of its residence time as a plasma membrane-bound protein, it is able to be used as a targeting agent for neurons. The target neurons include postganglionic sympathetic neurons and all noradrenergic and adrenergic neurons of the central nervous system. As such anti-DBH-SAP can be used for the production of acute peripheral autonomic failure in a model for the human disease. It can also be used for the analysis of the behavioral effects of noradrenergic or adrenergic ablation. When injected intraventricularly, noradrenergic neurons of the locus coeruleus are eliminated (Wiley & Lappi, 1994). The antibody and immunotoxin undergo retrograde transport.

      Entrez Gene Number
Entrez Gene SummaryThe protein encoded by this gene is an oxidoreductase belonging to the copper type II, ascorbate-dependent monooxygenase family. It is present in the synaptic vesicles of postganglionic sympathetic neurons and converts dopamine to norepinephrine. It exists in both soluble and membrane-bound forms, depending on the absence or presence, respectively, of a signal peptide.

      UniProt Number
UniProt SummaryFUNCTION: SwissProt: P09172 # Conversion of dopamine to noradrenaline.
COFACTOR: Binds 1 PQQ per subunit. & Binds 2 copper ions per subunit.
SIZE: 617 amino acids; 69065 Da
SUBUNIT: Homotetramer composed of two non-covalently bound disulfide-linked dimers.
SUBCELLULAR LOCATION: Soluble dopamine beta-hydroxylase: Cytoplasmic vesicle, secretory vesicle lumen. Cytoplasmic vesicle, secretory vesicle, chromaffin granule lumen. & Cytoplasmic vesicle, secretory vesicle membrane; Single-pass type II membrane protein. Cytoplasmic vesicle, secretory vesicle, chromaffin granule membrane; Single- pass type II membrane protein (Potential).
DISEASE: SwissProt: P09172 # Defects in DBH are the cause of DBH deficiency [MIM:223360]; also called norepinephrine deficiency or noradrenaline deficiency. This disorder is characterized by profound deficits in autonomic and cardiovascular function, but apparently only subtle signs, if any, of central nervous system dysfunction.
SIMILARITY: SwissProt: P09172 ## Belongs to the copper type II ascorbate-dependent monooxygenase family. & Contains 1 DOMON domain.
