      The pTriEx™-2 Hygro vector is designed to allow rapid characterization of target genes in multiple expression systems, and to allow rapid selection of stable transfected mammalian cells expressing high levels of target gene. With this vector a single recombinant plasmid can be used to test expression in E. coli, insect and vertebrate cells. Expression in vertebrate cells is mediated by a hybrid promoter composed of the CMV immediate early enhancer fused to the chicken β-actin promoter. The drug selection marker is expressed under the control of the EMC virus Cap-Independent Translation Enhancer (CITE) sequence (or IRES), allowing rapid selection of transfected mammalian cells using hygromycin. For expression in insect cells, pTriEx-2 Hygro contains flanking baculovirus sequences to permit the generation of recombinant baculoviruses using the BacVector® System. In baculovirus-infected insect cells, expression is driven by the very late p10 promoter. Expression in E. coli is regulated by the tightly controlled T7lac promoter. Expression can be induced in hosts such as NovaBlue by infecting with λCE6, a phage that constitutively expresses T7 RNA polymerase from the <λpL and λlpI promoters. Alternatively, pTriEx recombinant plasmids can be transferred into a (DE3)pLacI host that allows induction with IPTG.


      The pTriEx vector is covered by US Patent 6,589,783 owned by EMD Millipore Corporation or its Affiliates. This Product is sold for research use only. Any commercial use of the Product requires a commercial license from EMD Millipore Corporation. Commercial use shall include but not be limited to (1) use of the Product or its components in manufacturing; (2) use of the Product or its components to provide a service, information, or data to others in exchange for consideration; (3) use of the Product or its components (or any derivatives thereof) for therapeutic, diagnostic or prophylactic purposes (including as part of a device, chip, assay or other product); or (4) resale of the Product or its components, whether or not such Product or its components are resold for use in research. Further, the pTriEx vector allows for expression of target genes under control of the T7 promoter and is also covered by US Patent 5,693,489. This patent is licensed by EMD Millipore Corporation from Brookhaven National Labs for research use only. Commercial entities need to obtain a research use license from Brookhaven National Labs prior to use of this Product. Nothing contained herein shall be deemed to represent or warrant that additional third party rights are not required for use of this Product.
