Background InformationKiller cell immunoglobulin-like receptor 2DL1 (UniProt: P43626; also known as CD158 antigen-like family member A, Natural killer-associated transcript 1, NKAT-1, p58 natural killer cell receptor clones CL-42/47.11, p58 NK receptor CL-42/47.11, p58.1 MHC class-I-specific NK receptor, CD158a) is encoded by the KIR2DL1 (also known as CD158A, NKAT1) gene (Gene ID: 3802) in human. Killer-cell immunoglobulin-like receptors (KIRs) are cell surface glycoproteins predominantly expressed on Natural Killer (NK) Cells. They are shown to interact with MHC class I molecules to control cell lysis. KIR2DL1 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 22-245), a transmembrane domain (aa 246-264), and a cytoplasmic domain (aa 265-348). It also contains tow Ig-like C2-type domains (aa 42-107 and 142-205). It is expressed on NK cells and serves as a receptor for some HLA-C alleles such as w4 and w6. It is shown to Inhibit the activity of NK cells and prevent cell lysis. Two isoforms of KIR2DL1 have been described that are produced by alternative splicing. (Ref.: Meazza, R., et al. (2022). HLA. 100(2); 119-132; Falco, M., et al. (2010). J. Immunol. 185(1); 433-441).
