      Background InformationAS160 (Akt Substrate of 160 kDa, Akt1S1), Rab GAP (GTPase activating protein), is phosphorylated on multiple sites by Akt in response to insulin in adipocytes and contraction in muscles. AS160 is the long sought after link between the upstream insulin signaling through insulin/PI3K/Akt and GLUT4 vessicle translocation to the cell membrane. It has 6 Akt phosphorylation consensus sequences that are believed to become phosphorylated upon insulin stimulation. This phosphorylation is required for translocation of GLUT4-containing vesicles to the cell membrane1. A mutation of two or more of these phosphorylation sites results in a dramatic decrease in AS160 activity and insulin-stimulated GLUT4 redistribution at the cell membrane2. AS160 has also shown to be important for GLUT4 expcytosis2.

      Western Blot Analysis:
      A 1:1000-1:5000 dilution of this lot detected AS160 (Rab-GAP) in RIPA lysates from 3T3/L1 cells differentiated into adipocytes by MDI treatment. This lot also detects AS160 (Rab-GAP) in HEK293 lysates.
