      OverviewAll ChIPAb+ antibodies are individually validated for chromatin precipitation, every lot, every time. Each ChIPAb+ antibody set includes control primers (tested every lot by qPCR) to biologically validate your IP results in a locus-specific context. The qPCR protocol and primer sequences are provided, allowing researchers to validate ChIP protocols when using our antibody in their chromatin context. Each set also includes a negative control antibody to ensure specificity of the ChIP reaction.
      The ChIPAb+ PCAF validated antibody and primer set includes a PCAF antibody, a Normal Rabbit IgG, and positive control primers which amplify 127 bp of human PRAME The PCAF antibody and negative control IgG are supplied in a scalable "per ChIP" reaction size and can be used to functionally validate the precipitation of PCAF associated chromatin.

      Background InformationP/CAF(sometimes called KAT2B) is a transcription cofactor with a histone acetyltransferase (HAT) activity. Consistent with this protein playing a direct role in transcriptional regulation, P/CAF can acetylate core histones H3 and H4, chromatin proteins HMG17 and HMG I(Y), as well as human immunodeficiency virus Tat, transcription factor MyoD, and general transcription factors TFIIE and TFIIF. A role for P/CAF in cell growth and differentiation is suggested by its interaction in vivo and in vitro with p300/CBP, two large nuclear proteins that bind to sequence-specific factors involved in cell growth/differentiation. A role for P/CAF in cell division is also suggested by data showing that this protein competes with E1A for binding sites in p300/CBP to counteract E1A's mitogenic activity.

      Sonicated chromatin prepared from K562 cells (~4e6 cell equivalents per IP) were subjected to chromatin immunoprecipitation using 0.7 µg of either Normal Rabbit IgG (Part No. PP64B), or 0.7 µg Anti-PCAF (Part No. CS216159) and the Magna ChIP™ HiSens (Cat. # 17-10460). Successful immunoprecipitation of PCAF associated DNA fragments was verified by qPCR using ChIP Primers, PRAME (Part No. CS216150) as a positive locus, and ChIP Primers, SNRPN (Part No. CS207347) as a negative locus.
