      Background InformationSushi domain-containing protein 2 (UniProt: Q9UGT4; also known as SUSD2) is encoded by the SUSD2 gene (Gene ID: 56241) in human. SUSD2 is a single-pass type I membrane protein that serves as a tumor suppressor. It is synthesized with a signal peptide (aa 1-27), which is subsequently cleaved off to produce the mature form that contains a large extracellular domain (aa 28-783), a transmembrane domain (aa 786-806), and a short cytoplasmic domain (aa 807-822). Its sushi domain is localized to amino acids 723-780. SUSD2 also contains several domains that are frequently found in molecules implicated in cell-cell and cell-matrix adhesion and may play a role in regulating cell migration and/or invasion. SUSD2 is shown to be highly expressed in breast cancer, but shows restricted expression in normal tissues. Overexpression of SUSD2 in MDA-MB-231 cells has been linked to increased invasion, tumorigenesis, and angiogenesis. However, in high-grade serous ovarian carcinomas (HGSOCs) that does not depend on angiogenesis for metastasis, SUSD2 is shown to have a tumor suppressive effect. In HGSCOs it delays migration, epithelial-to-mesenchymal transition, and mesothelial clearance. Because SUSD2 is glycosylated and post-translationally cleaved, in Western blotting three bands may be detected that correspond to glycosylated, full-length protein; full-length SUSD2 with minimal glycosylation; and a band representing cleaved product of SUSD2.
