Trade Name
  • Chemicon
DescriptionThrombopoietin
OverviewTPO (Thrombopoietin, Mpl-ligand, MGDF) stimulates the proliferation and maturation of megakaryocytes and promotes increased circulating levels of platelets in vivo. Chemicon's Human TPO is a soluble 18.5 kDa protein containing 174 amino acid residues which comprises the receptor binding domain of the Mpl-ligand protein.

SourceE.coli
Specific ActivityThe ED50 was determined by the dose-dependent stimulation of the proliferation of M07e cells is <=1.0 ng/mL, corresponding to a specific activity of >=1x10(e6) units/mg.
Entrez Gene Number
Entrez Gene SummaryMegakaryocytopoiesis is the cellular development process that leads to platelet production. The protein encoded by this gene is a humoral growth factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis. This protein is the ligand for MLP/C_MPL, the product of myeloproliferative leukemia virus oncogene.

UniProt Number
UniProt SummaryFUNCTION: SwissProt: P40225 # Lineage-specific cytokine affecting the proliferation and maturation of megakaryocytes from their committed progenitor cells. It acts at a late stage of megakaryocyte development. It may be the major physiological regulator of circulating platelets.
SIZE: 353 amino acids; 37823 Da
SUBCELLULAR LOCATION: Secreted.
DOMAIN: SwissProt: P40225 Two-domain structure with an erythropoietin-like N- terminal and a Ser/Pro/Thr-rich C-terminal.
DISEASE: SwissProt: P40225 # Defects in THPO are a cause of essential thrombocythemia (ET) [MIM:187950]. ET is inherited as an autosomal dominant trait which is characterized by elevated platelet levels due to sustained proliferation of megakaryocytes, and frequently lead to thrombotic and haemorrhagic complications.
SIMILARITY: Belongs to the EPO/TPO family.

  • Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage ConditionsThe lyophilized powder, though stable at room temperature, is best stored at -20°C. Reconstitute in 5 mM Tris, pH 8.0 to a concentration of 0.5-1.0 mg/mL. This solution can be diluted into other aqueous buffers and stored at 4ºC for 1 week or -20ºC for future use. Reconstituted TPO should be stored in working aliquots at -20°C.
