OverviewThe SMC® Human IFNλ1 High Sensitivity Immunoassay Kit contains all reagents required to perform the assay. This assay takes advantage of a standard immunoassay workflow configured in a standard plate format. Assay protocols are similar to existing sandwich ELISA methods with two key differences: 1) Elution buffer disrupts the sandwich, separating the labeled detection antibody for quantification. 2) The SMCxPRO® immunoassay systems detect analytes using SMC® technology.
Background InformationInterferon lambda 1 (IFN-λ1), also know as IL-29, is a type of cytokine belonging to the interferon lambda family, which also includes IL-28A, IL-28B, and IL-29. Application NotesThe SMC® Human IFN-λ1 High Sensitivity Immunoassay uses a quantitative fluorescent sandwich immunoassay technique to measure IFN-λ1 in Human serum or plasma samples. A capture antibody specific for Human IFN-λ1 has been pre-coated onto paramagnetic microparticles (beads). The user pipettes beads, standards, and samples into uncoated microplate wells. During incubation, the IFN-λ1 present in the sample binds to the capture antibody on the coated beads. Unbound molecules are washed away during the subsequent wash steps. Fluor-labeled detection antibody is added to each well and incubated. This detection antibody recognizes and binds to IFN-λ1 that has been captured onto the beads, thus completing the sandwich. Elution buffer is added to dissociate the protein sandwich, releasing the fluor-labeled antibodies. The eluted antibodies are transferred to a SMC® 384-well Read Plate. The plate is loaded into the SMCxPRO® System where the labeled molecules are detected and counted. The number of fluor-labeled detection antibodies counted is directly proportional to the amount of IFN-λ1 present in the sample. The amount of IFN-λ1 in unknown samples is interpolated from a standard curve.

Cross ReactivityThis assay was tested for cross reactivity with IFN-ε, IFN-α2, IFN-γ, IFN-λ3, IFN-β, IFN-ω, and IFN-λ2 and no cross-reactivity was observed.
Accuracy
  • Spike recovery ranged between 61%-106% with a mean of 85% for serum and between 71% -120% with a mean of 95% for plasma.
