A cell-permeable, internally-quenched fluorogenic peptide substrate that is useful for detecting in vivo caspase-1-like protease activity in living cells or tissues. Efficiently cleaved by caspase-1 in vitro at the peptide bond between Asp and Ala residues (kcat/Km ~ 8.5 x 10-2 µM-1s-1) with an increase in fluorescence intensity (~525 nm) when excited at 380 nm. Both time-dependent and sub-cellular localization of caspase-1-like activity could be visualized in glutamate-induced rat hippocampal cell death.
