Application CommentsCan be used to localize α-synuclein in the central nervous system. For frozen sections, tissue should be fixed in 4% paraformaldehyde and 0.4% picric acid in 0.16 M sodium phosphate buffer, pH 6.9. Antibody should be titrated for optimal results in individual systems.

BackgroundThe major component of the vascular and plaque amyloid deposits in Alzheimer's disease (AD) is the amyloid β peptide. Non-A β component of Alzheimer's disease amyloid (NAC) is the second component in the amyloid plaques from brain tissues of AD patients. Its precursor protein (NACP) is now known to be identical to α-synuclein, a presynaptic protein in the human brain. In AD, synuclein/NAC proteins are found in presynaptic cholinergic nerve terminals that degenerate early in AD and are closely linked to β amyloid fibrils in senile plaques. α-synuclein is a highly conserved protein that shows 95% homology with rat synuclein 1 and good homology with Torpedo synuclein from the electric organ synapse and bovine phosphoneuroprotein 14 (PNP-14), a brain specific protein present in synapses. Recently α-synuclein has been linked to some cases of familial Parkinson's disease (PD), localizing in Lewy bodies, hallmark lesions of degenerating neurons in the brains of patients with PD. A missense mutation in the gene encoding α-synuclein gene has been identified in some of these PD patients.
