Background InformationParathyroid hormone-related protein (UniProt P12272), also known as Parathyroid hormone-like related protein, Parathyroid hormone-like hormone preproprotein, PLP, PTH-related protein, PTH-rP, PTHrP) is encoded by the PTHLH (also known as BDE2, HHM, PTHR, PTHRP) gene (Entrez Gene ID 5744) in human. Parathyroid hormone-related protein is initially translated as a 177 amino acid protein, which is then cleaved into PTHrP(1-34), PTHrP(1-36), PTHrP(38-94), and Osteostatin (also known as PTHrP (107-139)). Malignancy-associated hypercalcemia is caused by excessive production of parathyroid hormone-related protein (PTHrP) by the tumor. Neutralizing PTHrP(1-34) by antibody injection is shown to reduce serum calcium levels in hypercalcemic nude mice bearing transplanted human PTHrP-producing tumors, leading to their prolonged survival.
