Background InformationPeptidyl-prolyl cis-trans isomerase FKBP4 (EC 5.2.1.8; UniProt P27124; also known as 52 kDa FK506-binding protein, 52 kDa FKBP, 52 kDa immunophilin, FK506-binding protein 4, FKBP-4, FKBP-52, FKBP59, HBI, HSP-binding immunophilin, Immunophilin FKBP52, PPIase FKBP4, p59, Rotamase) is encoded by the FKBP4 (also known as P59) gene (Gene ID 100009148) in rabbit. PPIase FKBP4 (FKBP52, FKBP59, p59) is an immunophilin that binds the immunosuppressant drugs FK506 and rapamycin. FKBP59 was originally identified in the rabbit as a component of the unliganded, nontransformed steroid hormone receptor oligomeric complexes, where it was found associated with the chaperon protein hsp90 involved in protein folding. All the immunophilins are implicated in protein conformation regulations by catalyzing the cis-trans isomerization of proline imidic peptide bonds via their peptidylprolyl isomerase (rotamase) activity. FKBP59 is expressed at a higher level in the testis than in other organs. A high FKBP59 expression occurs in germ cells that have reached the same developmental stages either during prepubertal development or differentiation along the seminiferous tubules in the adult. The developmental stage-specific expression of FKBP59 during male germ cell differentiation suggests a role during the cell division.

Applications
ApplicationThis Anti-PPIase FKBP4 Antibody, clone KN382/EC1 is validated for use in Western Blotting, Immunocytochemistry, Immunohistochemistry, Immunoprecipitation and Radioimmunoassay for the detection of PPIase FKBP4.
Key Applications
  • Western Blotting
  • Immunocytochemistry
  • Immunohistochemistry
  • Immunoprecipitation
  • Radioimmunoassay
Application NotesWestern Blotting Analysis: 0.5 µg/mL from a representative lot detected PPIase FKBP4 in 10 µg of human brain tissue lysate.
Immunocytochemistry Analysis: A representative lot detected predominantly cytosolic immunoreactivity in human JAR choriocarcinoma cells by fluorescent immunocytochemistry (Sanchez, E.R., et al. (1990). Biochemistry. 29(21):5145-5152).
Immunohistochemistry Analysis: A representative lot stained both the cytosol and nucleus of epithelial and stromal cells using frozen uterus sections from estradiol-stimulated rabbits, while predominant nuclear staining was seen when using paraffin-embedded sections (Gasc, J.M., et al. (1990). Exp. Cell Res. 186(2):362-367).
Immunohistochemistry Analysis: A representative lot detected predominantly cytoplasmic PPIase FKBP4 (FKBP59) immunoreactivity in the early spermatids (eS), while both cytosolic and nuclear staining was seen in the midpachytene spermatocytes using paraformaldehyde-fixed, paraplast-embedded rabbit testis sections corresponding to stage VIII of the seminiferous epithelium cycle (Sananes, N., et al. (1998). Biol. Reprod. 58(2):353-360).
Western Blotting Analysis: A representative lot detected p59 in both the cytosolic and nuclear fractions of uterus extracts from estradiol-stimulated rabbits (Gasc, J.M., et al. (1990). Exp. Cell Res. 186(2):362-367).
