Background InformationHomeobox protein Nkx-2.1 (UniProt: P23441; also known as Thyroid nuclear factor 1, Thyroid transcription factor 1, TTF-1) is encoded by the Nkx2 (also known as Nkx-2.1, Titf1, Ttf-1, Ttf1) gene (Gene ID: 25628) in rat. Nkx-2.1 is a nuclear protein that is involved in the development and function of the brain, lungs, and thyroid gland. It serves as a transcription factor that binds and activates the promoter of thyroid specific genes such as thyroglobulin, thyroperoxidase, and thyrotropin receptor. It recognizes and binds to specific DNA sequences via a 60-amino acid DNA-binding domain called homeodomain. It is reported to be crucial in the maintenance of the thyroid differentiation phenotype. Its expression is restricted to regions of the developing brain within the diencephalon, in parts of the hypothalamus and neurohypophysis, and in the telencephalon. Its expression is shown to oscillate in a diurnal and melatonin-dependent manner in the preoptic area region in the hypothalamus, with maximal expression attained during the dark phase of the light/dark cycle. Nkx-2.1 is a highly selective marker for pulmonary adenocarcinoma and is a highly sensitive and specific immunomarker for distinguishing metastatic pulmonary from extrapulmonary adenocarcinoma.
