Background InformationMutated melanoma-associated antigen 1 (MUM1), also known as Protein expandere and PWWP domain-containing protein MUM1. MUM1’s normal role is as a DNA and chromatin associated protein that is involved in the cell’s response to DNA damage. It is driven to DNA breaks by its interaction with another DNA chromatin protein, TP53BP1, which it binds. The binding facilitates chromatin relaxation and entry of DNA repair enzymes. MUM1 is required for efficient DNA repair and cellular survival after DNA damage. Interestingly MUM1 was first identified by cloning the chromosomal breakpoints of a multiple myeloma cell line and was identified as a member of the interferon response transcriptional factor gene family (IFR). MUM1 appears to play a critical role in cell proliferation, differentiation and survival of various cancers, as well as monocytes and dendritic cells. MUM1 is expressed in B cells and plasma cells as well and in some populations of T lymphocytes. EMD-Millipore’s Anti-MUM1 mouse monoclonal antibody has been tested in against recombinantly expressed human MUM1 protein and in paraffin embedded immunohistochemistry on human melanoma, non-Hodgkin’s lymphoma and normal uterus tissue sections.
