Background InformationKallikrein-8 (EC 3.4.21.118; UniProt O60259; also known as hK8, hNP, Neuropsin, NP, Ovasin, Serine protease 19, Serine protease TADG-14, Tumor-associated differentially expressed gene 14 protein) is encoded by the KLK8 (also known as NP, NRPN, PRSS19, TADG14) gene (ORF UNQ283/PRO322; Gene ID 11202) in human. Kallikrein-8 is a member of the kallikrein subfamily of serine proteases, members of which are involved in diverse physiological functions and many are used as biomarkers for cancer. Kallikrein-8 is initially produced as a preproprotein with a signal (aa 1-28) and a propeptide (aa 29-32), cleavage of which releases the mature protein (aa 33-260). Kallikrein-8 and six other kallikreins (5, 6, 7, 10, 11, 13, and 14) are co-expressed in the outermost epidermal layer stratum corneum (SC), hair follicles, sebaceous and sweat glands, with kallikrein-8 being the most abundant trypsin-like kallikrein in SC and sweat. In addition to its involvement in epidermal homeostasis and wound healing, kallikrein-8 is also found to mediate the entry of human papillomaviruses (HPVs) into host cells.

Application NotesImmunohistochemistry Analysis: A 1:50 dilution from a representative lot detected Kallikrein 8/KLK8 in human cerebellum, mouse cerebellum, human kidney, and human pancreas.
ELISA Analysis: A representative lot was employed as the capture antibody for the detection of human LPL in patients-derived ovarian tumor cytosolic extracts by sandwich ELISA (Borgono, C.A., et al. (2006). Clin. Cancer Res. 12(5):1487-1493).
Activity Assay: A representative lot was employed to capture/isolate Kallikrein 8 (KLK8) from human sweat samples and epidermis stratum corneum (SC) extracts, as well as recombinant mature human KLK8 for activity assay (Eissa, A., et al. (2011). J. Biol. Chem. 286(1): 687–706).
