Background InformationGRB2-associated-binding protein 1 (UniProt: Q13480; also known as GRB2-associated binder 1, Growth factor receptor bound protein 2-associated protein 1, Gab1) is encoded by the GAB1 gene (Gene ID: 2549) in human. Gab1 is a signaling adaptor protein that acts as a platform for intracellular signal transduction triggered by activated receptor-type kinases and is involved in growth, transformation, and apoptosis. It plays a role in FGFR1, EGFR, and insulin receptor signaling and is shown to be phosphorylated on tyrosine residues in response to FGFR1, EGFR, and insulin receptor activation. These phosphorylations allow it to interact with several proteins that contain SH2 domains. Some of these phosphorylation sites serve as binding sites for phosphoatidylinositol 3 kinase (PI3K), Grb2, PLC gamma 1, Nck, and SHP2. Phosphorylations of Gab1 on tyrosine 627 and 659 are shown to be is critical for its binding to SHP2, and for activation of the ERK/MAPK pathway in response to EGF. Gab1 is also reported to be a critical component in the interaction between focal adhesion kinase and Met in cancer cell invasion. Mutations in GAB1 gene have been linked to autosomal recessive deafness that is characterized by prelingual, severe to profound hearing loss. (Ref.: Mouradian, M., et al. (2014).
