Background InformationSodium- and chloride-dependent GABA transporter 3 (UniProt: P31647; also known as GAT-3, Solute carrier family 6 member 11, GABA Transporter 3) is encoded by the Slc6a11 (also known as Gabt3, Gat-3, Gat-b) gene (Gene ID: 79213) in rat. GABA is reported to be the main inhibitory neurotransmitter in the mammalian brain, particularly in the basal ganglia network. Following its release from the presynaptic terminal, it is rapidly cleared from the extracellular space by GABA transporters (GATs) resulting it s the termination of inhibitory synaptic transmission. Thus far, four different GATs have been described in literature, GAT-1, GAT-2, GAT-3, and the Betain/GABA transporter type 1. GAT-3 is a multi-pass membrane protein that has one extracellular domain (aa 147-220), two cytoplasmic domains (aa 1-53 and 567-627), and twelve helical domains. It is shown to be highly expressed in the glomerular layer of the olfactory bulb, the inner nucleus of the retina, the thalamic paraventricular nucleus, and the globus pallidus. It terminates the action of GABA by its high affinity sodium-dependent reuptake into presynaptic terminals. The GABA-transporting action of GATs is dependent on the Na+ gradient across the membrane.
