Background InformationAlpha-(1,3)-fucosyltransferase 3 (FucT-III; Galactoside 3(4)-L-fucosyltransferase) is highly expressed in the small intestine, colon, stomach, and kidney. It is a member of the glycosyltransferase family of proteins which are involved in a glycosylation pathway that attaches fucose to Lewis epitopes of target molecules. This type of post-translational modification allows modified (fucosylated) molecules to recognize specific target substrates. Fucosyltransferases may therefore play an important role in communication at the cell surface. FucT-III may play a role in blood group determination as it is involved in the expression of antigens such as Lewis B, sialy Lewis X, Vim-2, and Lewis A.

Entrez Gene Number
Entrez Gene SummaryThe Lewis histo-blood group system comprises a set of fucosylated glycosphingolipids that are synthesized by exocrine epithelial cells and circulate in body fluids. The glycosphingolipids function in embryogenesis, tissue differentiation, tumor metastasis, inflammation, and bacterial adhesion. They are secondarily absorbed to red blood cells giving rise to their Lewis phenotype. This gene is a member of the fucosyltransferase family, which catalyzes the addition of fucose to precursor polysaccharides in the last step of Lewis antigen biosynthesis. It encodes an enzyme with alpha(1,3)-fucosyltransferase and alpha(1,4)-fucosyltransferase activities. Mutations in this gene are responsible for the majority of Lewis antigen-negative phenotypes. Multiple alternatively spliced variants, encoding the same protein, have been found for this gene. [provided by RefSeq, Jul 2008].

UniProt Number
UniProt SummaryFUNCTION: May catalyze alpha-1,3 and alpha-1,4 glycosidic linkages involved in the expression of Vim-2, Lewis A, Lewis B, sialyl Lewis X and Lewis X/SSEA-1 antigens. May be involved in blood group Lewis determination; Lewis-positive (Le+) individuals have an active enzyme while Lewis-negative (Le-) individuals have an inactive enzyme. Also acts on the corresponding 1,4-galactosyl derivative, forming 1,3-L-fucosyl links.

CATALYTIC ACTIVITY: GDP-beta-L-fucose + beta-D-galactosyl-(1->3)-N-acetyl-D-glucosaminyl-R = GDP + beta-D-galactosyl-(1->3)-(alpha-L-fucosyl-(1->4))-N-acetyl-beta-D-glucosaminyl-R.

PATHWAY: Protein modification; protein glycosylation.

SUBCELLULAR LOCATION: Golgi apparatus › Golgi stack membrane; Single-pass type II membrane protein. Note: Membrane-bound form in trans cisternae of Golgi.

TISSUE SPECIFICITY: Highly expressed in stomach, colon, small intestine, lung and kidney and to a lesser extent in salivary gland, bladder, uterus and liver.

SEQUENCE SIMLARITIES: Belongs to the glycosyltransferase 10 family.
Molecular Weight~50 kDa observed. The calculated molecular weight of this protein is 42 kDa, but can be observed at ~50 kDa due to high glycosylation. (Christensen, L, et al. (2000). Glycobiology. 10(9):931-939.)
