DescriptionUseful as a substrate for ELISA; acts as a hydrogen donor in peroxidase reactions. Radical cation has an absorption maximum at 414 nm.

Recommended reaction conditions
For Use as ELISA Substrate Note: Each tablet contains 5 mg and yields 5 ml ABTS® working substrate for ELISA when reconstituted as outlined below. 1. Dissolve ABTS at a final conc. of 1 mg/ml in 0.1 M sodium citrate buffer at pH 4.2. 2. Add 1.8 µl 30% H2O2 to 6 ml of the 1 mg/ml ABTS solution above (or 3 µl 3% H2O2 to 1 ml of the ABTS solution). 3. Add 1 ng HRP-conjugated antibody or HRP to 1 ml of the ABTS solution containing H2O2. 4. Incubate for 30 min to 1 h at 37°C (room temp is also OK, but the reaction is faster at 37°C). 5. Stop the reaction at the end of incubation with 1 ml (i.e. equal volume) of stop solution (0.625 M oxalic acid) and read the absorbance at 405 nm.
