New ready-to-use cleaning solution for chromatography resinsIssue: November 20, 2013

Chromatography is a process step which is performed with high value resins that require sanitization between batches, as well as prior to storage. While NaOH, the most widely used sanitant in bioprocessing, is very effective for polymer based resins, silica based resins require sanitization and storage with acidic solutions. Merck is pleased to now offer a ready-to-use, rapidly sanitizing PAB solution, specifically designed for the cleaning and storage of our ProSep® and Eshmuno® Protein A affinity chromatography media. This new stable PAB solution consists of benzyl alcohol, acetic acid and phosphoric acid, a composition which is effective against microbial contamination while protecting the valuable resins from decomposition at the same time. To prove the effectiveness of our PAB solution, we conducted a study in which the microbial kill effectiveness of PAB solution was compared to that of two concentrations of NaOH solution. The results indicated that PAB and NaOH demonstrated similar kill kinetics, however, NaOH was significantly less effective, even after several hours of exposure.
