OverviewTwo forms of Apo B exist: Apo B-100 and Apo B-48. The first is found in VLDL and LDL and is produced by the liver. The second is found in chylomicrons and originates in the intestine.

Apo B is purified from LDL isolated by density gradient ultracentrifugation (1.03-1.05 g/mL density range) followed by size exclusion chromatography in 10 mM deoxycholate, 50 mM NaCl, 50 mM Na2CO3, pH 10 (Walsh & Atkinson 1983).

PROPERTIES:

Apo B, which has a Mr of 550 kD, is the major protein found in LDL and is responsible for the cellular uptake of LDL particles from the plasma (Goldstein & Brown 1982). The Apo B prepared by this procedure is aggregated. The aggregates are not retained by a 0.45 mm filter, but migrate only a short distance into a 5% SDS PAGE gel. In the isolation buffer, Apo B gives a stable solution that is slightly hazy.

It occurs in plasma as two main isoforms, apoB-48 and apoB-100: the former is synthesized exclusively in the gut and the latter in the liver. The intestinal and the hepatic forms of apoB are encoded by a single gene from a single, very long mRNA. The two isoforms share a common N-terminal sequence. The shorter apoB-48 protein is produced after RNA editing of the apoB-100 transcript at residue 2180 (CAA->UAA), resulting in the creation of a stop codon, and early translation termination.

Heterozygotes for FHBL are usually asymptomatic with LDL cholesterol and apoB- 100 concentrations less than 50% of those in normal plasma. Homozygotes have extremely low plasma LDL cholesterol and apoB-100 concentrations, and clinical presentation may vary from no symptoms to severe gastrointestinal and neurological dysfunction similar to abetalipoproteinemia [MIM:200100]. & Defects in APOB are a cause of familial ligand-defective apolipoprotein B-100 (FDB) [MIM:144010]. FDB is a dominantly inherited disorder of lipoprotein metabolism leading to hypercholesterolemia and increased proneness to coronary artery disease (CAD). Storage ConditionsMaintain lyophilized material at <= -20°C for up to six months. Reconstitute to 1 mL by adding deionized water. If the deoxycholate is removed, Apo B-100 will slowly aggregate and become insoluble. After reconstitution maintain at +2-8°C for up to 4 weeks.
