Background InformationPartitioning defective 3 homolog (UniProt: Q99NH2; also known as PAR-3, PARD-3, Atypical PKC isotype-specific-interacting protein, ASIP, Ephrin-interacting protein, PHIP) is encoded by the Pard3 (also known as Par3) gene (Gene ID: 93742) in murine species. PAR-3 is an adapter protein that is involved in asymmetrical cell division and cell polarization processes. It plays a vital role in the formation of epithelial tight junctions and targets PTEN to cell junctions. Its association with PAR-6 may prevent the interaction of PAR-3 with F11 receptor/junctional adhesion molecule (F11R/JAM1), thereby preventing tight junction assembly. The PAR-3-PAR-6 complex also links GTP-bound Rho to atypical protein kinase C. PAR-3 is shown to be essential for establishment of neuronal polarity and normal axon formation. It is also involved in Schwann cell peripheral myelination where its deacetylation by Sirt2 reduces the activity of the polarity complex signaling component aPKC. In premyelinating Schwann cells, PAR-3 interacts with p75NTR through its PDZ1 binding domain to re-localize this receptor to axon-glial junctions, thereby promoting in vitro myelination. PAR-3 phosphorylation at serine 824 by PKC and PKC is shown to occur at the most apical tip of epithelial cell-cell contacts during the initial phase of tight junction formation. EGF-induced phosphorylation of tyrosine 1123 mediates its dissociation from (LIM Domain Kinase 2 ( LIMK2). Five isoforms of PAR-3 have been described that are produced by alternative splicing. All isoforms are expressed in the heart; however, its expression in brain is mainly limited to isoform 1. (Ref.: Beirowski, B., et al. (2011). Proc. Natl. Acad. Sci.
