DescriptionAnti-Orexin-B Antibody
Background InformationSeveral peptides associated with feeding behavior have been reported recently. Orexins (Orexin-A and Orexin-B) are a family of hypothalamic neuropeptides selectively expressed in the hypothalamus (Sakurai et al., 1998). Orexin-A and Orexin-B are derived from the same precursor (Prepro-orexin) by proteolytic cleavage. Prepro-orexin is an 130 aa peptide with a putative 33 aa secretory sequence, a hydrophobic core followed by residues with small polar side chains. The expression was detected in brain and to a small extent in testis (Sakurai et al., 1998; DeLecca et al., 1998). These neuropeptides bind and activate two closely related Orexin receptors--G protein coupled receptors (GPCRs) OX1R and OX2R.

FormatAffinity Purified
PresentationAffinity purified immunoglobulin. Liquid in PBS, with 0.1% BSA .
Quality LevelMQ100
Applications
ApplicationAnti-Orexin-B Antibody detects level of Orexin-B & has been published & validated for use in ELISA & WB.
Key Applications
  • ELISA
  • Western Blotting
Application NotesWestern blot: 1-10 μg/mL using Chemiluminescence technique

ELISA: 1:10,000 - 100,000 using 50-100 ng of control peptide per well.

Optimal working dilutions must be determined by end user.
Biological Information
ImmunogenA 13 amino acid peptide mapping near the amino terminus of mouse Orexin-B. The immunogen peptide has 92% similarity with human Orexin-B.
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostRabbit
SpecificityMouse/Rat Orexin-B. Reacts with full length Orexin-B. The AB3100 epitope is not found in Orexin-A. Since Orexins-A and Orexin-B are derived from an unprocessed pre-pro orexins (130 aa), the anti-Orexin-B antibodies may also react with pre-pro orexins.

Entrez Gene Number
Entrez Gene SummaryThis gene encodes a hypothalamic neuropeptide precusor protein that gives rise to two mature neuropeptides, orexin A and orexin B, by proteolytic processing. Orexin A and orexin B, which bind to orphan G-protein coupled receptors HCRTR1 and HCRTR2, function in the regulation of sleep and arousal. This neuropeptide arrangement may also play a role in feeding behavior, metabolism, and homeostasis.

UniProt Number
UniProt SummaryFUNCTION: SwissProt: O43612 # Neuropeptides that play a significant role in the regulation of food intake and sleep-wakefulness, possibly by coordinating the complex behavioral and physiologic responses of these complementary homeostatic functions. A broader role in the homeostatic regulation of energy metabolism, autonomic function, hormonal balance and the regulation of body fluids, is also suggested. Orexin-A binds to both OX1R and OX2R with a high affinity, whereas orexin-B binds only to OX2R with a similar high affinity.
SIZE: 131 amino acids; 13363 Da
SUBCELLULAR LOCATION: Rough endoplasmic reticulum (By similarity). Cytoplasmic vesicle (By similarity). Cell junction, synapse (By similarity). Note=Associated with perikaryal rough endoplasmic reticulum as well as cytoplasmic large granular vesicles at synapses (By similarity).
TISSUE SPECIFICITY: Abundantly expressed in subthalamic nucleus but undetectable in other brain regions tested (hypothalamus was not tested) and in heart, placenta, lung, liver, skeletal muscle, kidney and pancreas.
PTM: Specific enzymatic cleavages at paired basic residues yield the different active peptides.
DISEASE: SwissProt: O43612 # Defects in HCRT are a cause of narcolepsy [MIM:161400]. Narcolepsy is a neurological disabling sleep disorder, characterized by excessive daytime sleepiness, sleep fragmentation, symptoms of abnormal rapid-eye-movement (REM) sleep, such as cataplexy, hypnagogic hallucinations, and sleep paralysis. Cataplexy is a sudden loss of muscle tone triggered by emotions, which is the most valuable clinical feature used to diagnose narcolepsy. Human narcolepsy is associated with a deficient orexin system. Orexins are absent and/or greatly diminished in the brain and cerebrospinal fluid (CSF) of most narcoleptic patients. Human narcolepsy is primarily a sporadically occurring disorder but familial clustering has been observed.
SIMILARITY: SwissProt: O43612 ## Belongs to the orexin family.
