Application CommentsWill displace 50-70% of LDL binding to the receptor. For immunoblotting, include 2 mM Ca+2 with the antibody and avoid EDTA. This antibody has been reported to bind best to the unreduced LDL-R (see specificity application reference). Antibody should be titrated for optimal results in individual systems.

DescriptionPurified mouse monoclonal antibody generated by immunizing BALB/c mice with the specified immunogen and fusing splenocytes with Sp2/0-Ag14 myeloma cells (see application references). Recognizes the ~160 kDa LDL Receptor protein.
BackgroundThe low density lipoprotein receptor is responsible for mediating the uptake of LDL, a cholesterol carrying plasma lipoprotein. The receptor is a member of a class of cell surface proteins which are concentrated in clathrin coated pits and carry external ligands into cells by receptor-mediated endocytosis. The purified receptor is a glycoprotein with a molecular weight of 160,000. It is synthesized as a precursor of molecular weight 120,000 that is then glycosylated to the mature form by addition of N-linked and O-linked sugars in the Golgi apparatus. Unlike other hepatic lectins, the orientation of the LDL-R is such that the amino terminus is to the exterior of the cell. It has recently been determined that the LDL-R has sequence homology with the epidermal growth factor receptor precursor.
