Background InformationFanconi anemia group I protein (UniProt Q9NVI1; also known as Protein FACI) is encoded by the FANCI (also known as KIAA1794) gene (Gene ID 55215) in human. It plays a vital role in the repair of DNA double-strand breaks by homologous recombination and in the repair of interstrand DNA cross-links (ICLs) by promoting FANCD2 monoubiquitination by FANCL and participating in recruitment to DNA repair sites. It is also required for maintenance of chromosomal stability. FACI is reported to participate in S phase and G2 phase checkpoint activation upon DNA damage. It is monoubiquitinated by FANCL on Lys-523 during S phase and upon genotoxic stress and is deubiquitinated by USP1 as cells enter G2/M or once the DNA repair is completed. Mutations in the FANCI gene are known to cause Fanconi anemia, a developmental and cancer-predisposition syndrome. Ref: Smogorzewska A., et al. (2007) Cell 129, 289-301.

Western Blotting Analysis: 1.4 µg/mL from a representative lot detected endogenous FANCI in FANCD2-deficient EUFA1421 (kindly provided by Dr. Johan de Winter) cells. Exogenous FANCD2 expression stabilized FANCI and monoubiquitinated FANCI was detectable upoon 24-hour 60 ng/ml mitomycin C (MMC) treatment of FANCD2 transfectants (Courtesy of Martin Cohn, Ph.D., Department of Biochemistry, University of Oxford, UK).
