Product Information
FormatAffinity Purified
Control
  • Included free of charge with the antibody is 40 μg of control antigen (lyophilized powder). The stock solution of the antigen can be made up using 100 μL of sterile deionized water. For negative control, preincubate 1 μg of peptide with 10 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.
PresentationAffinity purified immunoglobulin. Lyophilized from PBS, pH 7.4, containing 1% BSA and 0.05% sodium azide as a preservative. Reconstitute with 200 μL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).
Quality LevelMQ100
Applications
ApplicationDetect Cav2.2 using this Anti-Cav2.2 Antibody validated for use in IC, IP & WB.
Key Applications
  • Immunocytochemistry
  • Immunoprecipitation
  • Western Blotting
  • Immunohistochemistry (Tissue)
Application NotesIn preparing full-length protein samples for Western blot or immunoprecipitation, the procedures should be performed at 4°C, with the following protease inhibitor mixture: pepstatin A (1 μg/mL), leupeptin (1 μg/mL), aprotinin (1 μg/mL), Pefabloc SC (0.2 mM), benzamidine (0.1 mg/mL), and calpain inhibitors I and II (8 μg/mL each).

Immunoprecipitation

Immunocytochemistry. Recommend subsequent staining with a labeled second antibody.

Western blot: 1.5 to 3.0 μg/mL (1:100-1:200) using ECL

Dilutions should be made using a carrier protein such as BSA (1-3%)

Optimal working dilutions must be determined by the end user.
Biological Information
ImmunogenPurified peptide CNB1 from 851-867 of alpha 1B subunit of rat brain voltage-gated calcium channel (Accession Q02294; Westenbroek et al. 1992) containing N-terminal lysine and tyrosine, conjugated to KLH with gluteraldehyde (Harlow & Lane 1988).
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.
HostRabbit
SpecificityRecognizes a 1B subunit (both LMW and HMW forms) of voltage-gated calcium channel. Does not cross react with any other calcium channel antigens tested so far.

SPECIES REACTIVITIES: It is expected that the antibody will also work on mouse due to sequence homology (15/17). Other species have not been tested.

Antibody TypePolyclonal Antibody
Entrez Gene Number
Entrez Gene SummaryVoltage-dependent Ca(2+) channels are multisubunit complexes found in the membrane of many excitable cells that regulate calcium entry (see MIM 601011). N-type calcium channels, which control neurotransmitter release from neurons, are dihydropyridine-insensitive and omega-conotoxin-sensitive. The alpha-1 subunit forms the pore through which calcium enters the cell, and is encoded by a family of at least 5 genes.[supplied by OMIM]

UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q00975 # Voltage-sensitive calcium channels (VSCC) mediate the entry of calcium ions into excitable cells and are also involved in a variety of calcium-dependent processes, including muscle contraction, hormone or neurotransmitter release, gene expression, cell motility, cell division and cell death. The isoform alpha-1B gives rise to N-type calcium currents. N-type calcium channels belong to the high-voltage activated (HVA) group and are blocked by omega-conotoxin-GVIA (omega-CTx-GVIA) and by omega-agatoxin- IIIA (omega-Aga-IIIA). They are however insensitive to dihydropyridines (DHP), and omega-agatoxin-IVA (omega-Aga-IVA). Calcium channels containing alpha-1B subunit may play a role in directed migration of immature neurons.
SIZE: 2339 amino acids; 262496 Da
SUBUNIT: Multisubunit complex consisting of alpha-1, alpha-2, beta and delta subunits in a 1:1:1:1 ratio. The channel activity is directed by the pore-forming and voltage-sensitive alpha-1 subunit. In many cases, this subunit is sufficient to generate voltage-sensitive calcium channel activity. The auxiliary subunits beta and alpha-2/delta linked by a disulfide bridge regulate the channel activity. Interacts with RIMS1 and RIMBP2 (By similarity).
SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.
TISSUE SPECIFICITY: Isoform Alpha-1b-1 and isoform Alpha-1b-2 are expressed in the central nervous system, but not in skeletal muscle or aorta.
DOMAIN: SwissProt: Q00975 Each of the four internal repeats contains five hydrophobic transmembrane segments (S1, S2, S3, S5, S6) and one positively charged transmembrane segment (S4). S4 segments probably represent the voltage-sensor and are characterized by a series of positively charged amino acids at every third position.
PTM: Phosphorylated in vitro by CaM-kinase II, PKA, PKC and CGPK (By similarity).
SIMILARITY: Belongs to the calcium channel alpha-1 subunit family. & Contains 1 EF-hand domain.
