Background InformationBcl-xL is a ~28 kDa member of Bcl-2 family of proteins and an important regulator of apoptosis. Bcl-xL forms heterodimers with BAX, BAK, and Bcl-2, and its overexpression in tumor cells confers resistance against chemotherapeutic drugs. Bcl-xL is phosphorylated on many sites including serine 62, a critical site for Bcl-xL response to microtubule-damaging drugs such as taxol and vinblastine. Phosphorylation of serine 62 (thought to be mediated by Jun N-terminal stress kinase (JNK) signaling) negatively regulates the anti-apoptotic function of Bcl-xL and controls the growth of neoplastic cells.

Entrez Gene SummaryThe protein encoded by this gene is a member of the BCL-2 family. BCL-2 family members are known to be regulators of programmed cell death. This protein positively regulates cell apoptosis by forming heterodimers with BCL-xL and BCL-2, and reversing their death repressor activity. Proapoptotic activity of this protein is regulated through its phosphorylation. Protein kinases AKT and MAP kinase, as well as protein phosphatase calcineurin were found to be involved in the regulation of this protein. Alternative splicing of this gene results in two transcript variants which encode the same isoform.

This interaction then facilitates the phosphorylation at Ser-118, a site within the BH3 motif, leading to the release of Bcl-X(L) and the promotion of cell survival. Ser-99 is the major site of AKT/PKB phosphorylation, Ser-118 the major site of protein kinase A (CAPK) phosphorylation (By similarity).

Quality AssurancePurified from rabbit serum by sequential epitope-specific chromatography. The antibody has been negatively preadsorbed using a non-phosphopeptide corresponding to the site of phosphorylation to remove antibody that is reactive with non-phosphorylated Bcl-xL. The final product is generated by affinity chromatography using a Bcl-xL-derived peptide that is phosphorylated at serine 62.
