Application CommentsFor immunocytochemistry, colcemid-arrested HeLa cells were fixed with 1% paraformaldehyde. Also detects histone H3 in Tetrahymena micronuclei. The antibody has been reported to detect mitotic cells in Xenopus, Caenorhabditis elegans, and Drosophila embryos. Does not cross-react with unphosphorylated histone H3. This antibody has also been reported to work for immunohistochemistry. Variables associated with assay conditions will dictate the optimal working dilution.

BackgroundHistone H1 and H3 phosphorylations correlate with the process of chromosome condensation. H3 mRNA-positive cells are most often found in tissues with a high proliferation rate, such as basal cells of the skin and crypt cells of the intestine.
