Brand Family Chemicon®
Trade Name
  • Chemicon
DescriptionAnti-TrkB Antibody
Alternate Names
  • Tyrosine kinase receptor B
Background InformationThe mammalian Trk family of receptor tyrosine kinases consists of three principle transmembrane proteins. The receptors can be activated by one or more of four neurotrophins - NGF, BDNF, NT3 and NT4/5. The TrkB family of transmembrane proteins serves as receptors for BDNF and NT-4/5 and these neurotrophins regulate biological responses such as cell survivial, proliferation and differentiation in normal and neoplastic neuronal cells.

Applications
ApplicationAnti-TrkB Antibody is an antibody against TrkB for use in IH.
Key Applications
  • Immunohistochemistry
Application NotesImmunohistochemistry: 1:3,000. The antibody has been used successfully on rat dorsal root ganglion and trigeminal ganglia in the rat spinal cord (cryostat free-floating sections). Suggested fixative is 4% paraformaldehyde.

Not suitable for Western blot.

Optimal working dilutions must be determined by end user.
Biological Information
ImmunogenExtracellular domain of the recombinant mouse receptor expressed in mammalian cells.
HostRabbit
SpecificitytrkB [BDNF/NT-4/5 growth factor receptor; tyrosine kinase receptor B]. The immunogen sequence was chosen to avoid cross reactivity with trkA or trkC.
Species Reactivity
  • Mouse
  • Rat
Species Reactivity NoteOther species have not yet been tested. It is expected to react with a wide range of mammalian species, based on the high sequence similarity.
Antibody TypePolyclonal Antibody
Entrez Gene Number
Entrez Gene SummaryThis gene encodes a member of the neurotrophic tyrosine receptor kinase (NTRK) family. This kinase is a membrane-bound receptor that, upon neurotrophin binding, phosphorylates itself and members of the MAPK pathway. Signalling through this kinase leads to cell differentiation. Mutations in this gene have been associated with obesity and mood disorders. Alternate transcriptional splice variants encoding different isoforms have been found for this gene.
Gene Symbol
  • NTRK2
  • Trk-B
  • TRKB
  • GP145-TrkB
  • EC 2.7.10.1

UniProt Number
UniProt SummaryFUNCTION: SwissProt: Q16620 # Receptor for brain-derived neurotrophic factor (BDNF), neurotrophin-3 and neurotrophin-4/5 but not nerve growth factor (NGF). Involved in the development and/or maintenance of the nervous system. This is a tyrosine-protein kinase receptor. Known substrates for the TRK receptors are SHC1, PI-3 kinase, and PLC- gamma-1.
SIZE: 822 amino acids; 91999 Da
SUBUNIT: Exists in a dynamic equilibrium between monomeric (low affinity) and dimeric (high affinity) structures. Binds SH2B2. Interacts with SQSTM1 (By similarity).
SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein.
TISSUE SPECIFICITY: Isoform TrkB is widely expressed, mainly in the nervous tissue. In the CNS, expression is observed in the cerebral cortex, hippocampus, thalamus, choroid plexus, granular layer of the cerebellum, brain stem, and spinal cord. In the peripheral nervous system, it is expressed in many cranial ganglia, the ophtalmic nerve, the vestibular system, multiple facial structures, the submaxillary glands, and dorsal root ganglia. Isoform TrkB-T1 is expressed in multiple tissues, mainly in brain, pancreas, kidney and heart. Isoform TrkB-T-Shc is predominantly expressed in brain.
PTM: Ligand-mediated auto-phosphorylation.
SIMILARITY: SwissProt: Q16620 ## Belongs to the protein kinase superfamily. Tyr protein kinase family. Insulin receptor subfamily. & Contains 2 Ig-like C2-type (immunoglobulin-like) domains. & Contains 2 LRR (leucine-rich) repeats. & Contains 1 protein kinase domain.
