Background InformationMotor neuron and pancreas homeobox protein 1 (UniProt: P50219; also known as Homeobox protein HB9) is encoded by the MNX1 (also known as HLXB9) gene (Gene ID: 3110) in human. MNX1 is a transcription factor that is expressed in lymphoid and pancreatic tissues. Its DNA binding homeobox region is localized in amino acids 241-300. It recognizes and binds to the regulatory elements of target genes and negatively modulates transcription. It plays a role in establishing motor neuron identity, in concert with LIM domain transcription factor LMO4. It is involved in negatively modulating transcription of interneuron genes in motor neurons, acting, at least in part, by blocking regulatory sequence interactions of the ISL1-LHX3 complex. It is also involved in pancreas development and function and may play a role in pancreatic cell fate specification. MNX1 is reported to be abnormally overexpressed in cervical cancer and its high expression is correlated with poorer clinicopathologic characteristics. It accelerates cell cycle transition by transcriptionally downregulating p21. It is also a critical regulator for normal pancreas and loss-of-function mutations are known to cause neonatal diabetes. Mutations in MNX1 gene have been linked to Currarino syndrome, an autosomal disorder that is caused by dorsal-ventral patterning defects during embryonic development.
