Background InformationHistones are highly conserved proteins that serve as the structural scaffold for the organization of nuclear DNA into chromatin. The histones have an amino terminal tail, a globular domain, and a carboxy-terminal tail. The four core histones, H2A, H2B, H3 and H4 assemble into the octamer (2 molecules of each). Subsequently, 146 base pairs of DNA are wrapped in a sequence-independent manner around the octamer, forming the basic subunit of chromatin, the nucleosome. Histone H4 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail, H4 is involved with the structure of the nucleosomes of the 'beads on a string' structure.

Application NotesImmunoblot Analysis:
A 1:100,000 dilution of a previous lot detected unmodified recombinant Histone H4 protein (Catalog # 14-412) and Histone H4 in acid extracts from both untreated HeLa cells and cells treated with sodium butyrate (hyper-acetylated) or colcemid (hyper-phosphorylated) (Figure B).

Chromatin Immunoprecipitation:
An independent laboratory used a previous lot of this antibody in chromatin immunoprecipitation (Issac, C., 2006).

Multiplexing:
Serial dilutions of a previous lot were incubated with histone peptides containing various unmodified peptides spanning the aminoterminus of Histone H4 conjugated to Luminex® microspheres. The antibody recognized both modified and unmodified peptides.

Two molecules of each of the four core histones (H2A, H2B, H3, and H4) form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures. This gene is intronless and encodes a member of the histone H4 family. Transcripts from this gene lack polyA tails but instead contain a palindromic termination element. This gene is found in the large histone gene cluster on chromosome 6.
