      Application Data
Detection of human JNK1 and JNK2, phosphorylated on Tyr183 and Thr185, by immunoblotting. Samples: Whole cell lysate from HEK293 cells left untreated (lane 1) or treated with UV irradiation (lanes 2-6). Primary antibody: PhosphoDetect™ Anti-SAPK/JNK (pTyr183,pThr185) Rabbit pAb (Cat. No. 559309) (1:1000) pre-incubated without peptide (lanes 1,6) or with immunogen phosphopeptide (lane 2), a generic phosphothreonine-containing peptide (lane 3), a generic phosphotyrosine-containing peptide (lane 4), or non-phosphopeptide corresponding to the immunogen (lane 5). Detection: chemiluminescence.

DescriptionRabbit polyclonal antibody adsorbed against non-phosphopeptide corresponding to the immunogen phosphorylation site, followed by immunoaffinity chromatography. Recognizes the ~49 kDa JNK1/SAPK1 protein and the ~55 kDa JNK2/SAPK2 protein phosphorylated at Thr183 and Tyr185.
BackgroundC-Jun-N-terminal kinase (JNK), also called stress-activated protein kinase (SAPK) is a member of the mitogen activated protein kinase (MAPK) family. JNK is activated in response to various extracellular stimuli, including pro-inflammatory cytokines. Phosphorylation of threonine and tyrosine in the Thr-Pro-Tyr domain is required for full activation. Mitogen activated protein kinase kinase (MKK4) preferentially phosphorylates tyrosine 185, and MKK7 causes preferential phosphorylation of threonine 183.
