UGT1A9 is a single-pass membrane glycoprotein that belongs to the UDP-glycosyltransferase family. UDP-Glucuronosyltransferases are a major group of phase II drug metabolizing enzymes and each enzyme in this family displays a unique substrate specificity and tissue-specific expression patterns. UDP-Glucuronosyltransferases mediate the transfer of glucuronic acid from UDP-glucuronic acid to various aglycone substrates making them more hydrophilic and readily excretable. UGT1A9 is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to generate the mature form. It is involved in the conjugation and subsequent elimination of potentially toxic xenobiotics and endogenous compounds. Two isoforms of UGT1A9 have been described that are produced by alternative splicing. Both isoforms are expressed in liver and kidney. However, isoform 2 lacks the transferase activity and acts as a negative regulator of isoform 1. Clone 14G11 is shown to recognize both glycosylated and de-glycosylated forms of UGT1A9, however, the reactivity is shown to be higher in de-glycosylated form. (Ref.: Oda, S., et al. (2012).
