Background InformationC-X-C chemokine receptor type 3 (UniProt: O88410; also known as CXC-R3, CXCR-3, Interferon-inducible protein 10 receptor, IP-10 receptor, CD183) is encoded by the Cxcr3 (also known as Cmkar3) gene (Gene ID: 12766) in murine species.CXCR-3 is a multi-pass membrane protein of the G-protein coupled receptor family. It is highly expressed on effector Th1-type CD4+ and effector CD8+ cells and serves as a chemokine receptor for interferon-inducing ligands, CXCL9, CXCL10, and CXCL11. CXCR-3 is reported to play a critical role in the migration of T cells in the microenvironment of the peripheral tissue and lymphoid compartment, that facilitates the interaction of T cells with antigen presenting cells leading to the generation of effector and memory cells. CXCR-3 expression on effector T cells is shown to be essential for their entry into sites of inflammation. CXCR-3 is an N- glycosylated protein, which also undergoes sulfation on tyrosine 27 and 29. These sulfations are essential for its binding to CXCL10. Clone 173 can block receptor binding of CXCL10 and CXCL11, but not CXCL9 in murine cells. When injected into mice along with low dose of rapamycin, it is shown to prolong cardiac and islet allograft survival. (Ref.: Uppaluri, R., et al. (2008).
