      SynonymsARKRERTYSFGHHA
DescriptionAn optimal peptide substrate for assaying AKT/PKB/Rac-protein kinase activity in vitro. The peptide undergoes phosphorylation at the Ser site (Km = 3.9 µM). Reported to competitively inhibit histone H2B phospho-rylation (Ki = 12 µM) by AKT.
      FormOff-white solid
      FormulationSupplied as a trifluoroacetate salt.
Intert gas (Yes/No) Packaged under inert gas
      Recommended reaction conditions
The reaction mixture (30 µl) contains 20 mM HEPES, pH 7.4, 100 µM ATP with 3 µCi of [γ-32P]-ATP, 10 mM DTT, 10 mM MgCl2, 0.5 mM EGTA, and 50 µM AKTide-2T. Reaction begins with the addition of 0.1 µg of purified AKT. Incubation is carried out at 25°C for designated time periods (5 to 15 min). Reaction is terminated by adding 10 µl of stop solution containing 8 N HCl and 1 mM ATP. Aliquots are then spotted on P81-phosphocellulose paper, washed four times in 0.5% phosphoric acid, and quantified by scintillation counting. Appropriate blanks are run concurrently.
