      BackgroundInsulysin or insulin degrading enzyme (IDE; accession no. P14735), also known as insulinase, is evolutionarily conserved, ubiquitously expressed, ~110 kDa zinc metalloprotease. It is located in the cytosol, on the cell surface, and may also be secreted. IDE is the major enzyme responsible for degrading insulin and is, thus, critical for glucose, lipid, and protein metabolism, as well as for cell growth and differentiation. IDE also degrades cerebral Aβ, a protein whose cleaved peptides contribute to the pathology of Alzheimer's disease. Homozygous deletion of the IDE gene in mice results in an increase in the level of Aβ as high as 65%, as well as an increase of 180% in serum insulin levels accompanied by impaired glucose tolerance, which is a hallmark of type 2 diabetes mellitus (DM2). Although hyperinsulinemia and glucose intolerance are the hallmark of DM2, etiology of the disease and the specific role of IDE in this process is not well understood. In human genetic studies, the IDE region of chromosome 10q has shown linkage to late-onset AD, DM2, and fasting plasma glucose level. In a recent study IDE was also identified as a cellular receptor for chickenpox virus (VZV) glycoprotein E, gE, which is involved in viral entry and cell-to-cell spread.
