      Background InformationGlucagon is a hormone that is proteolytically cleaved into at least eight different functional peptides. It is produced by pancreatic cells and it has binding sites in the brain, intestines, and liver, among other tissues. Glucagon is secreted in response to hypoglycemia. Through the combined activation of Gsα- and Gq-coupled transmembrane receptors and their downstream signaling pathways, glucagon stimulates the release of glucose from the liver into the bloodstream. One important mechanism underlying this process is the activation of protein kinase A (PKA). PKA signals result in increased transcription of phosphoenolpyruvate carboxykinase, which encodes the rate-limiting enzyme in the production of glucose from noncarbohydrate substrates. PKA signals may also increase the breakdown of glycogen, and suppress glycolysis. Glucagon may also engage in calcium-dependent signaling via PLC. The importance of glucagon in regulating glucose homeostasis, in conjunction with insulin, is underscored by findings that glucagon and glucagon receptor signaling are disrupted in type II diabetes.
