      ApplicationImmunoblotting (5 µg/ml)
DescriptionPurified mouse monoclonal antibody (see application references). Recognizes the ~28 kDa cathepsin K protein.
BackgroundCathepsins are normal lysosomal proteases, which are expressed in all cells where they function in protein turnover. They have also been shown to be capable of degrading extracellular matrix proteins. Cathepsin K, also known as OC-2, is a cysteine endoproteinase that is predominantly expressed in osteoclasts and is structurally and functionally related to cathepsin L and cathepsin S. The cloned human cDNA encodes a protein of 329 amino acids showing a 94% homology to rabbit cathepsin K. Northern blot analysis show elevated levels of cathepsin K mRNA in osteoarthritic hip bones and very high levels in osteoclastoma compared to low levels in other tissues such as kidney, liver, spleen and lung. The predominant expression of cathepsin K in osteoclasts suggests an important role in human osteoclastic bone resorption and may provide possible rationale for the treatment of osteoporosis and certain forms of arthritis.
