DescriptionRabbit polyclonal antibody supplied as undiluted serum. Recognizes the ~53 kDa p53 phoshorylated at Ser15.
Backgroundp53 (393 amino acids) exhibits sequence-specific DNA binding, directly interacts with various cellular and viral proteins, and induces cell cycle arrest in response to DNA damage by inducing GADD45, WAF1, and MDM2. p53 is the most commonly mutated gene in human cancer with most mutations occurring in the DNA contact residues. p53 binds damaged DNA non-specifically via its C-terminal 75 amino acids which in turn may be responsible for subsequent activation, sequence-specific DNA binding, and WAF1 induction allowing DNA repair to take place. p53 down regulates bcl-2 expression and up regulates bax expression, but may not always be necessary for apoptosis. In addition to the afore mentioned proteins, two other proteins involved in replication and recombination, RPA and RAD51, also bind p53. Multi-site phosphorylation of p53 is key in mediating many of these events. Within its N-terminal domain, p53 is phosphorylated by kinases responsive to UV radiation, cytokines, DNA damage and growth factors. C-terminal phosphorylation is mediated by protein kinases involved in growth stimulation, cell cycle control and apoptosis. These C-terminal phosphorylation events are involved in controlling the specific DNA binding function of p53, and may play a role in regulating other functions of p53.
