OverviewThe Periodic acid solution 0.5% for the PAS reaction(periodic acid Schiff) for the detection of aldehyde and mucosubstances in microscopy, is a ready to use solution used for the detection of mucopolysaccharides, glycogen, muco- and glycoproteins, glyco-, phospholipids, basal membranes and collagen. The material is treated with periodic acid creating aldehyde groups which react with Schiff's reagent and give a bright red color reaction, thus the nuclei appear blue.
In the PAS reaction, the histological specimen material is first treated with periodic acid, resulting in the oxidation of the 1,2-glycols into aldehyde groups. The addition of Schiff’s reagent (fuchsin-sulfuric acid)(Cat.no. 109033) in the second step causes the aldehydes to react to form a brilliant red color. In the end result, the PAS reaction yields a specific color reaction with unsubstituted polysaccharides, neutral mucopolysaccharides, muco- and glycoproteins, and glyco- and phospholipids.
The PAS reaction can be further combined with the Alcian blue staining method to detect mucosubstances (glycosaminoglycans).

Anorganické kyseliny a ich anhydridy najprv zrieďte alebo hydrolizujte opatrným primiešaním do ľadovej vody a potom neutralizujte (rukavice, komora s odsávaním) hydroxidom sodným (kat. č. 105587). Pred vyliatím do nádoby D skontrolujte pH pomocou indikátorových prúžkov pH-Universal (kat č. 109535). Dymivú kyselinu sírovú opatrne primiešajte po kvapkách do 40 % kyseliny sírovej (kat. č. 109286). Majte po ruke veľké množstvo ľadu na chladenie. Po dostatočnom ochladení upravte vysokokoncentrovanú kyselinu sírovú tak, ako je uvedené vyššie. Obdobne anhydridy je možné premeniť na príslušné kyseliny. Kyslé plyny (napr. halidy vodíka, chlór, fosgén, oxid siričitý) je možné prebublať cez zriedený roztok hydroxidu sodného a po neutralizácii zneškodniť v nádobe D.
