Revision20-May-2008 RFH
SynonymsSulfobiotin-X-NHS, Sulfosuccinimidyl-6-(biotinamido)hexanoate
DescriptionWater solubility is extremely convenient, eliminating the need to dissolve the reagent in an organic solvent such as dimethylformamide (DMF) and dimethylsulfoxide (DMSO) prior to use.
FormOff white to tan solid
Recommended reaction conditions
Buffers containing amines (such as Tris or glycine) should be avoided because they can compete with the reaction. It is also very important that the stock solution be prepared just prior to addition of the reaction mixture, because these compounds are prone to hydrolysis by water. Water-soluble crosslinkers can be dissolved in water or buffer at a concentration of 10-25 mM. Phosphate buffers may precipitate sulfo-N-hydroxysuccinimide (sulfo-NHS) containing compounds. In addition, the solid crosslinker can be added directly to the sample to minimize hydrolysis. Hydrolysis is more pronounced in dilute protein solutions, but can be minimized by using more concentrated protein solution (greater than 5 mg/ml). If you prefer to work with stock solutions, we recommend that you dissolve the crosslinker in anhydrous DMSO at a concentration of 10-25 mM. This solution can then be diluted into the buffer of your choice just prior to use. General Handling 1. Store desiccated at 4°C. 2. Warm contents to room temperature completely before opening. Allow at least 30 min. 3. Wipe the vial to remove any moisture. 4. After opening, store under nitrogen. 5. Protect from moisture.
