DescriptionAnti-phospho-TBC1D1 Antibody (Ser237)
Alternate Names
  • TBC1 domain family member 1
  • TBC1 (tre-2/USP6, BUB2, cdc16) domain family, member 1
Background InformationTBC1 domain family member 1 (UniProt: Q86TI0; also known as TBC1D1) is encoded by the TBC1D1 gene (Gene ID: 23216) in human. TBC1D1 is a member of the TBC1 Rab-GTPase family of nuclear proteins that is highly expressed in skeletal muscle. Its major action is to regulate exercise-mediated glucose uptake into muscle. It is involved in the trafficking and translocation of GLUT4-containing vesicles and insulin-stimulated glucose uptake into cells. Insulin and muscle contraction are reported to increase TBC1D1 phosphorylation on phospho-Akt substrate motifs. Insulin-stimulated phosphorylation by AKT is shown to stimulate GLUT4 translocation. Following muscle exercise, phosphorylation of TBC1D1 at serine 237 by AMPK and 14-3-3 protein binding capacity are significantly increased. Knockdown of TBC1D1 in C2C12 muscle cells is shown to increase fatty acid oxidation and reduce muscle levels of GLUT4. Although TBC1D1 knockout mice do not exhibit any disruption in their whole-body insulin action, they display impaired exercise-regulated metabolism. A mutation in TBC1D1 (R125W) has been linked to obesity in human. (Ref.: Stockli, J., et al. (2015). Diabetes. 64(6); 1914-1922; An, D., et al. (2010). Diabetes. 59(6); 1358-1365; Frosig, C., et al. Applications
ApplicationAnti-phospho-TBC1D1 (Ser237), Cat. No. 07-2268, is a rabbit polyclonal antibody that detects TBC1D1 phosphorylated on serine 237 and is tested for use in Peptide Inhibition Assay and Western Blotting.
Key Applications
  • Western Blotting
  • Peptide Inhibition Assay
Application NotesTested Applications

Peptide Inhibition Assay: Target band detection in HEK293 cell lysate was prevented by preblocking of a representative lot with the immunogen phosphopeptide, but not the corresponding non-phosphopeptide.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
Biological Information
ImmunogenKLH-conjugated linear peptide corresponding to 13 amino acids surrounding phosphoserine 237 of human TBC1 domain family member 1 (TBC1D1).
EpitopePhosphorylated Ser237
ConcentrationPlease refer to the Certificate of Analysis for the lot-specific concentration.

Entrez Gene Number
Entrez Gene SummaryTBC1D1 is the founding member of a family of proteins sharing a 180- to 200-amino acid TBC domain presumed to have a role in regulating cell growth and differentiation. These proteins share significant homology with TRE2 (USP6; MIM 604334), yeast Bub2, and CDC16 (MIM 603461) (White et al., 2000 [PubMed 10965142]).[supplied by OMIM].

Quality AssuranceEvaluated by Western Blotting in lysate from HEK293 cells treated with phorbol-12-myristate-13-acetate
(PMA).

Western Blotting Analysis (WB): A 1:500 dilution of this antibody detected TBC1D1 lysate from HEK293 cells treated with phorbol-12-myristate-13-acetate (PMA). However, the detection was inhibited blots pre-incubated with protein phosphatase.
