DISEASE: SwissProt: P60201 # Defects in PLP1 are the cause of Pelizaeus-Merzbacher disease (PMD) [MIM:312080]. PMD is an X-linked neurologic disorder of myelin metabolism. It is characterized by early impairment of motor development (during the first three months of life) and, later, by the development of abnormal movements and progressive spastic paraplegia. & Defects in PLP1 are the cause of spastic paraplegia type 2 (SPG2) [MIM:312920]. SPG2 is a form of X-linked hereditary spastic paraplegia (HSP). HSP is a group of inherited degenerative spinal cord disorders characterized by a slow, gradual, progressive weakness and spasticity of the legs. Rate of progression and the severity of symptoms is quite variable. Initial symptoms may include difficulty with balance, weakness and stiffness in the legs, muscle spasms, and dragging the toes when walking. In some forms of the disorder, bladder symptoms (such as incontinence) may appear, or the weakness and stiffness may spread to other parts of the body. Additional features include nystagmus, dysarthria and mental retardation, that are found in the complicated form of spastic paraplegia.
