  • Neurexin 1-alpha
Background InformationNeurexin-1 (UniProt: Q9CS84; also known as Neurexin I-alpha, Neurexin-1-alpha) is encoded by the Nrxn1 (also known as Kiaa0578) gene (Gene ID: 18189) in murine species. Neurexins are a large family of proteins that act as neuronal cell-surface receptors. Their function appears to be isoform-specific. They are heavily glycosylated and processed neuronal transmembrane cell surface proteins that are involved in cell recognition and adhesion. Neurexin-1 (the isoform) is a single-pass type I presynaptic membrane glycoprotein that is synthesized with a signal peptide that is subsequently cleaved off to produce the mature form that contains an extracellular domain (aa 31-1438), a transmembrane domain (aa 1439-2459), and a cytoplasmic domain (aa 1460-1514). It has a long N-terminus with six laminin G-like domains and play an important role in synaptic signal transmission. It is involved in the regulation of calcium channel activity and Ca2+-triggered neurotransmitter release at synapses and at neuromuscular junctions. It binds to neuroligins through an extracellular laminin neurexin sex-hormone-binding protein (LNS) domain. The neuroligin neurexin complex, a heterophilic adhesion system, promotes assembly and maturation of synapses through bidirectional signaling. In this complex, postsynaptic neuroligins interact trans-synaptically with presynaptic neurexins. This pan-neurexin antibody recognizes - and β-neurexins. Seven isoforms of Neurexin-1 have been described that are produced by alternate promoter usage and alternative splicing. (Ref.: Taniguchi, H., et al. (2007). J.
