Background InformationMucins are a family of highly glycosylated proteins that are present on the surface of, and are secreted by, many glandular epithelial cells. Mucins provide a barrier between the luminal membranes of cells in the epithelium and their environment. For instance, in the digestive tract, the mucin barrier protects the epithelial cells from the action of proteolytic enzymes. These high molecular weight glycoproteins consist of a nonglobular protein backbone (apomucin) and many O-linked glycans. Salivary Mucin 5B proteins are some of the largest polypeptides, with molecular weights ranging from 14 - 40 x 103 kDa.

FormatPurified
PresentationPurified immunoglobulin. Liquid in PBS containing 0.1% sodium azide.
Quality LevelMQ100
Applications
ApplicationDetect Mucin 5B using this Anti-Mucin 5B Antibody, clone 19.4E validated for use in ELISA, WB, IH.
Key Applications
  • ELISA
  • Western Blotting
  • Immunohistochemistry
Application NotesWestern blot; Bands in the high molecular weight range of 200kDa or above are expected for highly glycosylated proteins.

Immunohistchemistry: Paraffin sections. Deglycosylating the tissue is critical to expose epitope. There are a number of deglycosylation protocols available and a discussion of these related to Mucin can be found in Cao, Y. et al. (1997)(see references).

ELISA

Optimal working dilutions must be determined by end user.

UniProt SummaryFUNCTION: SwissProt: Q9HC84 # Gel-forming mucin that is thought to contribute to the lubricating and viscoelastic properties of whole saliva and cervical mucus.
SIZE: 5703 amino acids; 590499 Da
SUBCELLULAR LOCATION: Secreted.
TISSUE SPECIFICITY: Expressed on surface airway epithelia. Expressed mainly in mucous cells of submucosal glands of airway tissues. Highly expressed in the sublingual gland. Also found in submaxillary glands, endocervix, gall bladder, and pancreas.
DOMAIN:SwissProt: Q9HC84 The cysteine residues in the Cys-rich subdomain repeats are not involved in disulfide bonding.
PTM: Highly glycosylated. C-, N- and O-gylcosylated. C- mannosylated in the Cys-rich subdomains probably on the first Trp residue of the WXXW motif. Highly O-glycosylated in the Ser/Thr- rich tandem repeat (TR) region. The repeat region is about 59% O- glycosylated with a high abundance of NeuAc(2)Hex(1)HexNac1-ol.
SIMILARITY: Contains 1 CTCK (C-terminal cystine knot-like) domain. & Contains 1 TIL (trypsin inhibitory-like) domain. & Contains 3 VWFC domains. & Contains 4 VWFD domains.
