Background InformationPhospholipid hydroperoxide glutathione peroxidase, mitochondrial (EC 1.11.1.12; UniProt O70325; also known as GPx-4, GSHPx-4, mtPHGPx, PHGPx, Phospholipid hydroperoxide glutathione peroxidase mitochondrial, snGPx, Sperm nuclei glutathione peroxidase) is encoded by the Gpx4 gene (Gene ID 625249) in murine species. Gpx4 is a member of the glutathione peroxidase (GPx) family of peroxidases that protect cells against oxidative damage by reducing lipid hydroperoxides to their corresponding alcohols and by reducing free hydrogen peroxide to water. Eight glutathione peroxidase members (GPx1-8) have been identified in human. Inducible Gpx4-knockout in transgenic mice reveals the essential role of Gpx4 in maintaining proper kidney function, and Gpx4 depletion results in massive cell death of renal tubular epithelia in vivo. While the most abundant mammalian GPx, GPx1, that forms tetramers, GPx4 exists in the monomeric form. Three alternatively spliced GPx4 isoforms originate from the same Gpx4 gene, a cytosolic (also called the short form), a mitochondrial (also called the long form), and a nuclear form. The short form is expressed ubiquitously in embryonic and adult tissues in varying degrees, while the expression of the other two forms is restricted to testis where they exert significant functions during male gametogenesis.
