Background InformationGangliosides are sialic acid-containing glycosphingolipids that are most abundant in the nervous system. They contain one or more carbohydrate residues linked to a hydrophobic lipid moiety through a glycosidic linkage. Six major and many minor ganglioside species have been identified in mammalian brains. They have been identified as receptors for bacterial toxins and viruses and serve as tumor-associated antigens. In GM2, G refers to ganglioside, the M is for monosialic and 2 refers to the fact that it was the second monosialic ganglioside to be discovered. GM2 is a minor component of cell membranes that has been shown to accumulate in Tay-Sachs and Sandhoff diseases, that are caused by defects in the lysosomal glycosidases or specific co-activators, which results in accumulation of the substrates, such as glycosphingolipids, including gangliosides. Hydrolysis of GM2 is facilitated by the cooperation of two proteins, -hexosamidase A (HexA) and its cofactor GM2 activator protein (GM2AP). Clone GMB28 is shown to be highly specific for GM2 and GM2(NeuGc) and does not show reactivity with other gangliosides or neutral glycolipids. (Ref.: Kotani, M., et al. (1992). Biochim.
