Background InformationThe Non-Radioactive FoxO1 Transcription Factor Assay kit is provided in a 96-well format. During the assay, the Capture Probe, a double stranded biotinylated oligonucleotide containing the consensus sequence for FoxO1 binding (5'-TTGTTTAC-3'), is mixed with whole cell or nuclear extract in the Transcription Factor Assay Buffer provided directly in the Streptavidin coated plate. When incubated together, FoxO1 contained in the nuclear extract binds to its consensus sequence. The biotinylated double stranded oligonucleotide bound to FoxO1 protein is immobilized and any inactive, unbound material is washed away. The bound FoxO1 transcription factor is detected with a specific rabbit anti-FoxO1, primary antibody. An HRP-conjugated secondary antibody is then used for detection and provides sensitive colorimetric detection that can be read in a spectrophotometric plate reader. Included in the kit are positive control whole cell extract, a non-specific double stranded oligonucleotide, and a specific double stranded competitor oligonucleotide.

UniProt Number
UniProt SummaryFUNCTION: Transcription factor. Ref.9
SUBUNIT STRUCTURE: Interacts with LRPPRC By similarity.
SUBCELLULAR LOCATION: Cytoplasm. Nucleus. Note: Shuttles between cytoplasm and nucleus.
TISSUE SPECIFICITY: Ubiquitous.
PTM: Phosphorylated by AKT1; insulin-induced By similarity. IGF1 rapidly induces phosphorylation of Ser-256, Thr-24, and Ser-319. Phosphorylation of Ser-256 decreases DNA-binding activity and promotes the phosphorylation of Thr-24, and Ser-319, permitting phosphorylation of Ser-322 and Ser-325, probably by CK1, leading to nuclear exclusion and loss of function. Phosphorylation of Ser-329 is independent of IGF1 and leads to reduced function. Phosphorylated upon DNA damage, probably by ATM or ATR.
INVOLVEMENT IN DISEASE: Chromosomal aberrations involving FOXO1 are a cause of rhabdomyosarcoma 2 (RMS2) [MIM:268220]; also known as alveolar rhabdomyosarcoma. Translocation (2;13)(q35;q14) with PAX3; translocation t(1;13)(p36;q14) with PAX7. The resulting protein is a transcriptional activator.
SEQUENCE SIMILARITIES: Contains 1 fork-head DNA-binding domain.

Usage Statement
  • Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage ConditionsStable for 1 year at -20ºC from date of receipt.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
