DescriptionPurified mouse monoclonal antibody antibody generated by immunizing mice with the specified immunogen and fusing splenocytes with Sp2/0 mouse myeloma cells. Recognizes the E2F1 protein.
BackgroundE2F transcription activity is the result of regulation by a family of heterodimers that are encoded by distinct genes. The different members of the E2F family form complexes with the DP1 protein and it is this complex which is responsible for the regulatory activity. The gene for E2F1 was the first of the E2F family to be cloned and was shown to map to chromosome 20q11. The gene contains 7 exons and is regulated at the mRNA levels through changes in transcription. E2F1 has been shown to regulate both G1 and S phase cyclins and in particular there is evidence for a type of feedback regulatory loop between E2F and cyclin D1. Overexpression of E2F1 has been shown to lead to the induction of apoptosis possibly through the inhibition of cyclinD1 dependent kinase activity coupled with the induction of a p16 related transcript. In addition to the transcription level regulation of E2F1, there is also control of E2F1 protein levels by the ubiquitin-proteosome dependent degradation pathway. Ubiquitination is blocked by Rb stabilizing the E2F1 complex formation and directly controlling cell cycle progression.
